Type of Bioactive Compounds Against A-Glucosidase and Acetylcholinesterase from Edible Wild Mushroom

เห็ดได้มีการนำมาใช้เป็นส่วนประกอบของยาแผนโบราณในหลายประเทศโดยเฉพาะประเทศแถบเอเชีย นอกจากนี้ยังมีรายงานความหลากหลายของสารออกฤทธิ์ทางชีวภาพในเห็ดที่เกี่ยวข้องกับสุขภาพของมนุษย์
งานวิจัยนี้ศึกษาปริมาณสารออกฤทธิ์ทางชีวภาพในกลุ่มลิพิดจำนวน 2 ชนิดคือ โอลีเอไมด์และกรดเฮกซะเดคาโนอิกจากเห็ดป่ากินได้จำนวน 5 ชนิด ได้แก่ เห็ดตับเต่า (M1), เห็ดแครง (M2), เห็ดผึ้งแดงคราม (M3), เห็ดผึ้งหวาน (M4) และเห็ดผึ้งแดง (M5) รวมถึงศึกษาการยับยั้งเอนไซม์ที่เกี่ยวข้องกับโรคเบาหวานและโรคอัลไซเมอร์ของสารโอลีเอไมด์และกรดเฮกซะเดคาโนอิก ผลการทดลองพบว่าสารสกัดของเห็ด M2 และ M4 มีปริมาณสารโอลีเอไมด์และกรดเฮกซะเดคาโนอิกสูงที่สุด ดังนั้นสารโอลีเอ-ไมด์และกรดเฮกซะเดคาโนอิกจากสารสกัด M2 และ M4 ถูกทำให้บริสุทธิ์โดยใช้เทคนิคโครมาโตกราฟีแบบผิวบาง โดยใช้ระบบตัวทำละลายคลอโรฟอร์ม:เมทานอล (5:5 v/v) สำหรับสารสกัด M2 และเมทานอล:คลอโรฟอร์ม:น้ำ (51:42:7 v/v/v) สำหรับสารสกัด M4 ผลการทดลองพบว่าได้สารบริสุทธิ์โอลีเอไมด์ (M2P) จากสารสกัด M2 และสารบริสุทธิ์กรดเฮกซะเดคาโนอิก (M4P) จากสารสกัด M4 เมื่อศึกษาชนิดของการยับยั้งเอนไซม์แอลฟา-กลูโคซิเดส และ เอนไซม์แอซิติลโคลีนเอสเทอเรสของสารสกัด M2สารบริสุทธิ์ M2P และสารมาตรฐานโอลีเอไมด์ พบว่ามีชนิดของการยับยั้งเป็นแบบผสมทั้งสองเอนไซม์และเมื่อหาชนิดของการยับยั้งเอนไซม์แอลฟา-กลูโคซิเดส ของสารสกัด M4 สารบริสุทธิ์ M4P และสารมาตรฐานกรดเฮกซะเดคาโนอิก พบว่ามีการยับยั้งเป็นแบบผสม แบบแข่งขัน และแบบไม่แข่งขันตามลำดับ และเมื่อหาชนิดของการยับยั้งเอนไซม์แอซิติลโคลีนเอสเทอเรสของสารสกัด M4 สารบริสุทธิ์M4P และสารมาตรฐานกรดเฮกซะเดคาโนอิกพบว่ามีชนิดของการยับยั้งเป็นแบบผสม นอกจากนี้ยังพบว่าสารมาตรฐานโอลีเอไมด์ และกรดเฮกซะเดคาโนอิกมีการจับกับกรดอะมิโนที่บริเวณเร่งของเอนไซม์แอลฟา-กลูโคซิเดส และแอซิติลโคลีนเอสเทอเรส
ผลการทดลองแสดงให้เห็นว่าเห็ดเป็นแหล่งของสารออกฤทธิ์ทางชีวภาพที่สามารถยับยั้งเอนไซม์แอลฟา-กลูโคซิเดส และแอซิติลโคลีนเอสเทอเรสได้ และสามารถนำไปประยุกต์ใช้ในการรักษาโรคเบาหวานและโรคอัลไซเมอร์ต่อไป
Abstract
Edible wild mushrooms have been used as an ingredient in traditional medicinein many countries, particularly in Asia. Various bioactive compounds related to humanhealth have been reported in edible wild mushrooms.
This research investigated the content of two lipid-based bioactive compoundsoleamide and hexadecanoic acid from five species of edible wild mushrooms, including Phlebopus portentosus (Berk & Broome) Boedijn (M1), Schizophyllum commune Fr.(M2), Boletus aemilii Barbier (M3), Boletus edulis Bull (M4), and Heimiella japonica (Hongo) E. Horak (M5). Moreover, the research also investigated the inhibitory effects of oleamide and hexadecanoic acid against the enzymes related to diabetes mellitus (DM) and Alzheimer’s disease (AD). The results revealed that mushroom extracts M2 and M4 contained the highest amounts of oleamide and hexadecanoic acid. Therefore, oleamide and hexadecanoic acid were purified from M2 and M4 using Preparative Thin Layer Chromatography (PTLC) technique, with chloroform: methanol (v/v/v) for M4.
The results showed that purified oleamide (M2P) and hexadecanoic acid (M4P) were successfully purified from M2 and M4, respectively. The inhibition analysis revealed that the M2 extract, M2P, and standard oleamide exhibited mixed type inhibition against α-glucosidase and acetylcholinesterase (AChE) The M4 extract, M4P, and standard hexadecanoic acid demonstrated mixed-type, competitive, and non-competitive inhibition, respectively, against α-glucosidase. For AChE, the M4 extract, M4P, and standard hexadecanoic acid showed mixed-type inhibition. For more observations, the oleamide and hexadecanoic acid were located in the active site of α-glucosidase and AChE. The results suggest that edible wild mushrooms are the source of bioactive compounds capable of inhibiting α-glucosidase and AChE and may have potential applications in the treatment of diabetes mellitus and Alzheimer’s disease.
